Can I make a GUI cover the entire screen including the bar at the top that has the chat button and backpack button?
Is it possible to let a GUI cover the entire screen?

Okay, there are two ways this can go: change the topbar transparency to 0, or resize it. It's your choice. But let's break it down.
Topbar Solution
With this, you can place this local script either in ReplicatedFirst or StarterGui. What this essentially does is to make the bar opaque, meaning it isn't see-through anymore!
--LocalScript.
game:GetService('Players').LocalPlayer:WaitForChild('PlayerGui'):SetTopbarTransparency(0)
Size Solution
This is probably the more simpler one, but I will explain the positioning and why. When you do GUIs, most of the time, you are aiming for compatibility on all devices, hence you should use {x,0}{y,0} (the scales) rather than the second part of each brace, the studs.
Scales are out of 1, meaning if you set the scale to 1 for x and y, the entire screen will be covered. However, the topbar is actually above that, about a tenth of a screen above. Therefore, your position for the GUI should be around {0,0}{-.1,0} (You can lower the value to ensure). While after that, you might realize, that a tenth of the screen in showing on the bottom. To resolve this, we'll just do what we did, and add a tenth of a screen on the y-value. And since 1 + .1 = 1.1, the size should be {1,0}{1.1,0}.
Hope this clears up any confusion!

Change the frames properties under size to 1,0,1,0. I think that's what you want to do. If not tell me what you are trying to accomplish and I will try to help out.
No, including the bar at the top that has the chat and backpack buttons. When I do 1,0,1,0 it fills everything except the bar.
